Steady State

Pharmacokinetic condition in which drug input rate equals elimination rate — reached after ~5 half-lives of regular dosing.

DEFINITION

At steady state, the rate at which a drug enters the systemic circulation equals the rate at which it is eliminated, and plasma concentrations fluctuate within a stable range across dosing intervals.

For a drug with linear pharmacokinetics, steady state is reached after approximately five elimination half-lives of regular dosing. For once-weekly Semaglutide with a plasma half-life of ~1 week, this corresponds to 4–5 weeks of consistent dosing.
