Reconstitution is the aseptic laboratory procedure by which a lyophilised peptide is dissolved into a sterile diluent — most commonly bacteriostatic water for injection, sterile water for injection, or 0.9% sodium chloride — to prepare a stock solution at a known concentration.
Correct reconstitution requires accurate diluent-volume calculation from the target concentration, gentle swirling rather than vortexing to avoid foaming and shear-induced aggregation, and a brief equilibration period to allow full dissolution. Post-reconstitution solutions are typically stored at 2–8 °C or aliquoted and frozen at −20 °C.
Reconstitution converts a shelf-stable powder into a working solution whose concentration underpins every downstream dose, assay dilution and stability calculation.
