SURPASS Programme

Randomised trial programme evaluating Tirzepatide in adults with type 2 diabetes across monotherapy and active-comparator studies (SURPASS-1 through SURPASS-5).

DEFINITION

The SURPASS programme is a series of randomised phase-3 clinical trials evaluating once-weekly subcutaneous Tirzepatide in adults with type 2 diabetes. SURPASS-1 (Rosenstock 2021) tested Tirzepatide as monotherapy against placebo; SURPASS-2 (Frías 2021) was a head-to-head comparison with once-weekly Semaglutide 1 mg.

The programme extends across multiple background-therapy and active-comparator arms (SURPASS-3 through SURPASS-5). Primary and key secondary endpoints centre on HbA1c change from baseline and body-weight change.

WHY IT MATTERS

SURPASS is the core evidence base for Tirzepatide in type 2 diabetes and includes the first head-to-head trial versus a long-acting GLP-1 mono-agonist.
