DEFINITION
SNAC (sodium N-[8-(2-hydroxybenzoyl)amino]caprylate; salcaprozate sodium) is a small-molecule absorption enhancer developed for oral peptide delivery. It transiently raises local pH in the stomach, protects the co-formulated peptide from pepsin and increases membrane permeability enough to allow absorption of a small but pharmacologically sufficient fraction of the dose.
SNAC is the enabling excipient in oral Semaglutide (Rybelsus), the first oral GLP-1 receptor agonist product.
WHY IT MATTERS
SNAC is the reason an acylated GLP-1 peptide can be delivered orally at all.
