DIO Model

Also known as: Diet-induced obesity model

Rodent (typically mouse or rat) fed a high-fat diet to model obesity and metabolic dysregulation for preclinical research.

DEFINITION

The diet-induced obesity (DIO) model is the standard rodent preclinical platform for evaluating anti-obesity and metabolic pharmacology. Mice or rats are fed a defined high-fat diet, typically for 8–20 weeks, to induce adiposity, insulin resistance and dyslipidaemia.

DIO models are used extensively to benchmark GLP-1 receptor agonists such as Semaglutide alongside dual and triple incretin agonists in vivo.

WHY IT MATTERS

DIO is the workhorse preclinical model for GLP-1 receptor agonist and multi-agonist metabolic pharmacology.
